Effect of dietary supplementation level of vitamin A on lipid peroxide content and antioxidative system in ethanol-administered rat

Abstract
The present study was conducted to investigate the effect of dietary supplementation of vitamin A on lipid peroxide content and antioxidative system in ethanol-administered rat. Sprague-Dawley rats were given a liquid diet containing 36% of total calories as ethanol for 5 weeks. The pair-fed control rats received an isocaloric amount of diet containing sucrose instead of ethanol. Additionally, the liquid diet contained different levels of retinyl acetate (1A: 0.35, 2A: 1.75, 3A: 3.50 §·/§¤ liquid diet). Body weight gain and food efficiency ratio of ethanol groups were lower than those of pair-fed groups. On the other hand, the body weight gain of 2AE group was significantly higher than that of the other ethanol groups. Plasma triglyceride and total cholesterol were increased by ethanol feeding, while phospholipid content tended to be reduced in ethanol groups. And these levels were not changed by dietary supplementation of vitamin A. The levels of plasma and hepatic TBARS were increased after chronic ethanol feeding, but those of 2AE group were significantly reduced compared with the other ethanol groups. Hepatic superoxide dismutase activity was decreased by ethanol feeding. However, catalase activity was enhanced by chronic ethanol feeding, with no difference between 3AE and 3AP rats. Glutathione S-transferase and glutathione peroxidase activities were increased in ethanol groups, with the lowest activities in 2AE rats. These results suggest that ethanol feeding enhances lipid peroxidation and the appropriate level of vitamin A supplementation may reduce enhanced lipid peroxidation and related damage to some extent.
